We identified a region (-57 to -45) in the proximal promoter of CST1 that binds a specific protein, termed X-proximal-binding factor (XBF), present in embryonic CNS nuclear extracts. Using 5′ deletion mutants of the CST1 promoter, XBF was shown to bind to an X-box-like motif in the -57 to -45 region of the proximal promoter. The homeobox protein Hox-3.1 was shown to inhibit binding of XBF to the CST1 X-box, and to inhibit the in vivo expression of the CST1 gene in the developing CNS.
